Sirtuins are a family of seven evolutionarily conserved NAD+-dependent enzymes (SIRT1 through SIRT7) found in different cellular compartments. They regulate a vast array of biological processes including gene silencing, DNA repair, mitochondrial biogenesis, fatty acid oxidation, glucose homeostasis, and inflammation. SIRT1, the most studied mammalian sirtuin, deacetylates transcription factors including p53, FOXO, PGC-1alpha, and NF-kB, providing anti-aging, metabolic, and anti-inflammatory effects. Sirtuins are obligate NAD+ consumers, meaning their activity is directly limited by cellular NAD+ availability. The age-related decline in NAD+ levels therefore reduces sirtuin activity, contributing to the hallmarks of aging. This dependency makes NAD+ supplementation a compelling research strategy for supporting sirtuin function.
