Collagen synthesis is the complex multi-step biosynthetic process by which fibroblasts and other connective tissue cells produce collagen, the most abundant protein in the human body comprising approximately 30% of total protein mass. The process begins with gene transcription and translation of procollagen alpha chains, followed by post-translational modifications including hydroxylation of proline and lysine residues (requiring vitamin C as a cofactor), glycosylation, and triple-helix formation. Procollagen is then secreted extracellularly, where propeptides are cleaved and mature collagen molecules self-assemble into fibrils and fibres. GHK-Cu has been demonstrated to stimulate collagen types I, III, and V production by fibroblasts, while also increasing the production of metalloproteinases that break down damaged collagen for replacement. BPC-157 similarly supports collagen deposition at wound sites through growth factor modulation.
