Thymosin Beta-4 (TB4) is a 43-amino acid protein belonging to the beta-thymosin family, originally isolated from calf thymus tissue but now known to be expressed ubiquitously in nearly all nucleated cells. It is the primary intracellular G-actin-sequestering protein, maintaining a pool of monomeric actin available for rapid polymerisation when cells need to migrate, divide, or change shape. TB4 is released from platelets upon tissue injury and plays a critical role in the early phases of wound repair by promoting endothelial cell migration, keratinocyte migration, and angiogenesis. Beyond wound healing, TB4 has demonstrated anti-inflammatory properties by downregulating inflammatory cytokines, anti-fibrotic effects that reduce scar formation, and cardioprotective actions in models of myocardial infarction. TB-500 is a synthetic peptide based on the active region of TB4 that retains its key tissue-repair properties in a more pharmacologically manageable format.
